Overview

Colorectal cancer is a malignant tumor that originates in the epithelial lining of the colon or rectum, the terminal segments of the large intestine, and is one of the most frequently diagnosed cancers and a major cause of cancer mortality worldwide. Within the context of intestinal biology, the disease typically begins as benign adenomatous polyps that may progress to invasive carcinoma through the accumulation of genetic and epigenetic alterations affecting the intestinal mucosa. Risk factors include increasing age, family history and hereditary syndromes, chronic intestinal inflammation, and dietary and lifestyle influences, several of which are modifiable. Because precancerous lesions can be detected and removed, the disease is highly amenable to prevention and early intervention through screening with colonoscopy or stool-based tests such as fecal immunochemical testing. Treatment is stage-dependent and may combine surgical resection, chemotherapy, radiotherapy, and immunotherapy. Research relevant to intestinal disease emphasizes the cellular and molecular events in the colonic and rectal wall, screening and early detection, and the influence of diet and environment. Studies published in this area address gene regulation in colorectal cancer, mechanisms of apoptosis resistance in cancer cells, adhesion-molecule and gene-expression analyses in tumor tissue, anatomical features of the colon relevant to interval cancers, screening programs, dietary intervention, and the broader clinical and survivorship aspects of the disease.
